Abstract

Sirtuin 1 (SIRT1)-mediated peroxisome proliferator-activated receptor gamma coactivator 1 (PGC-1α) deacetylation is potentially a key step for inducing mitochondrial biogenesis. The polyphenol curcumin, derived from the rhizome Curcuma longa L., is a natural antioxidant that exhibits various pharmacological activities and therapeutic properties. However, the effect of nanocurcumin on skeletal muscle in the regulation of mitochondrial biogenesis remains unknown. The purpose of the present study was to examine the effects of endurance training (eTR) and nanocurcumin treatment on SIRT1, 5^ adenosine monophosphate-activated protein kinase (AMPK), PGC-1α, mitochondrial protein expression of cytochrome c oxidase subunit IV (COX-IV), in rat skeletal muscle. Ten-week-old male Wistar rats were randomly divided into non-eTR and eTR groups. curcumin and nanocurcumin (100 mg/kg-BW/day) give peroral in the non-eTR and eTR group animals for 28 days to investigate the effect of nanocurcumin alone and the combined effect of nanocurcumin with eTR. Western blotting (WB) was performed to detect the presence of proteins. Compared to eTR alone, nanocurcumin treatment combined with eTR increased the expression levels of mitochondrial marker protein COX-IV in the gastrocnemius. Furthermore, our result demonstrated that curcumin treatment together with eTR increased the phosphorylation of AMPK, PGC-1α Addition nanocurcumin treatment alone, in the absence of eTR, increased PGC-1α, COX-IV expression,. Taken together, these results suggest that the combination of nanocurcumin treatment and eTR has the potential to accelerate mitochondrial biogenesis in skeletal muscles via AMPK-PGC-1alpha signaling pathway.
